A Study on chemoprophylaxis using several anthelmintic drugs in clonorchiasis


Abstract
The author has investigated on the 3 anthelmintic drugs for the preventive use of clonorchiasis in rats.
The drugs are praziquantel, niclofolan and bithionol.
The metacercariae of Clonorchis sinensis were obtained from Pseudorasbora parva caught in Kim Hae and were isolated by the digestion technique.
The experimental animals were sacrificed in the period of 30th days after the infection with metacercariae of Clonorchis sinensis.
The results obtained were summarized as follows
1. In the groups which praziquantel 30mg, niclofolan 7n and bithionol 6.4n was administered at 10 minutes after infection with metacercariae, there were not found adult worms of Clonorchis sinensis in the biliary ductal system of the rats. But, when the number of infecting metacercariae was increased, or when the dosages of the drugs were reduced, the detections of the adult worms were positive in the groups niclofolan and bithionol.
And the detection rates of the both groups were somewhat increased in proportional to the 2 conditions described earlier.
While the group giving praziquantel was negative in spite of the dosage reduction to 1 / 6 of the first dose and infection with the increased number of metacercariae.
2. In the group giving praziquantel at 10 minute interval before infection with metaeercariae, the adult worm of Clonorchis sinensis was not found in the liver of the rats.
But the niclofolan administered group showed 0.1% rate of the detection and the bithionol group showed 0.5% rate of that. By the p mge of the time gradually, only in the groups giving bithio of at 30 minute and 60 minute intervals after infections with metacercariae, the adult worms of Clonorchis sinensis were found.
3. In the groups giving praziquantel at 10 minute interval before and after infections with metacercariae, both groups were all negative. And also the detections to the groups giving praziquantel at 8 hour interval before and after infections with metacercariae were negative.
But, there were positive detections in the groups with the longer intervals than 8 hour interval, and they were gradually increased with the passage of the time.
On the basis these results, it was suggested that praziquantel was effective to the chemoprophylaxis of clonorchiasis in rats, which was given at 8 hour intervals before and after infections with metacercariae and its dosage was reduced to 1 / 6 of the first dose.
